The PathHunter PTGFR Total Internalization eXpress kit provides an easy to-use cell based assay to study drug candidates (small molecules or biologics) affecting GPCR activation and internalization. This kit uses EFC technology to measure ligand-induced internalization of PTGFR via detection of localization to the early endosome. The included cell line overexpresses ProLink-tagged PTGFR and an EA-tagged early endosome protein. Upon ligand-induced receptor activation, resulting internalization of the PTGFR receptor to the early endosome leads to production of EFC signal. eXpress kits are a convenient, ready-to-use format which contains all materials needed to run the assay, including single-use vials of cryopreserved cells, cell plating media, detection reagent, and assay plates. Biotium CF488A and PI Apoptosis Assay Kit
Phosphatidylserine (PS) is translocated from the inner to the outer surface of the cell for phagocytic cell recognition. The human anticoagulant, Annexin V, is a 35 kDa Ca2 -dependent phospholipid protein with a high affinity for PS. Annexin V labeled with CF488A (Abs/Em= 490/515 nm) can identify apoptotic cells in green by binding to PS exposed on the outer leaflet. Our CF488A dye is superior to fluorescein/FITC as it is not affected by pH and has much better photostability. Propidium iodide (PI) is impermeant to live cells or early apoptotic cells, but stains necrotic cells and late apoptotic cells with red fluorescence (Abs/Em with DNA = 535/617 nm). Both PI and Annexin V-CF488A can be excited using the 488 nm line of a flow cytometer. The kit has reagents sufficent for 100 assays.
